a gram stain is usually done first no matter what if there is no idea and if it comes out variable or negative, an acid fast stain would definitively tell you its myco. tb. I would go with C since gram stains on acid fast organisms are inconclusive since they are variable or negative unless the stain is left on for a long time (like an hour)

c: an acid-fast stain They are impermeable to the usual stains, e.g. Gram's stain. They are known as "acid-fast bacilli" because of their lipid-rich cell walls, which are relatively impermeable to various basic dyes unless the dyes are combined with phenol. Once stained, the cells resist decolorization with acidified organic solvents and are therefore called "acid-fast". They could easily be stained by Ziehl-Neelsen stain
